Design: Solexa PCR were carried out in 40ul reactions containing 10ul purified PCR production,1ul Forward primers, 1ul Reverse primers , 20ul Phusion , 8ul ddH2O; PCR reactions were as follows: denaturation at 98 for 30s, followed by 10 cycles of 10s denaturation at 98 , annealing at 65 for 30 s, extension at 72 for 30s and a final 7-min extension at 72 .
